I do miss the venting, screened skylight window I had in the camper of my Tacoma, just a simple little window that let in light and kept it cooler. Toyota advises against having such a modification in the Sienna, but I have seen photos of far more extreme roof modifications than that little window (approximately a foot square).
I use my vehicle to travel to shows with my artwork, then as my short person's "home away from home;" so the pod-like aspect of the Sienna is an improvement over exiting the truck to enter the camper.
Does anyone have any comments or insights regarding my situation?

Do you mean a typical 14" square RV vent?  Those things are trash compared to automotive components (I know, I have them in my travel trailers), and there's no way I would cut a perfectly good vehicle's roof to install one.  My biggest concern would be sealing, although there's also appearance and finding a place to install it without cutting a structural reinforcement.

There are some venting accessories which temporarily install in the top of a door window opening; the window is partially open and closed up against them.

A real automotive sunroof would be a better bit of hardware, but they are not normally screened (and I understand the desire for a screen).

Well, I have the vent, but haven't had it installed although a reputable shop says it can be done. I had one in my aluminum camper for years that never leaked at all and only looked like a half bubble on the roof.
The issue of possibly cutting a structural reinforcement is significant. I also wonder if there are any wires or cables up there? I have seen photos of Siennas with the roof completely sheared off, one replaced with a pop-up roof, the other with a sliding window, sunroof, moonroof, whatever - it was big and it opened.
I have survived staying in the Sienna this spring and summer, but it would be so much more comfortable with that roof ventilation.

My 2011 Limited has front and rear sunroofs.  The rear one is huge.
